AUMsavercan be used to efficiently silence or regulate different RNA modalities, such as messenger RNA, micro-RNA and long non-coding RNA for in vitro studies. In addition, ASOs can be designed to act as steric blockers without eliciting RNase-H mediated degradation. AUMsaver provides potent knockdown of the target RNA with high sequence specificity.
AUMsaver use RNase H-mediated cleavage (Figure 1), as opposed to the RNAi pathway involving the RISC complex (which is prone to off target effects). RNAse H recognizes the ASO/RNA duplex and cleaves the RNA within the hybrid. Following cleavage, the fragmented RNA is further degraded by nucleases, whereas the ASOs are recycled within the cell.
